Dire Wolves Revived After 12,500 Years: A Breakthrough in De-Extinction Technology

Dallas-based biotech company Colossal Biosciences announced today that they have successfully birthed three dire wolf pups, marking a significant milestone in de-extinction technology. The dire wolf, scientifically known as Aenocyon dirus, has been extinct for approximately 12,500 years, having once roamed North America as a top predator during the Late Pleistocene era.

The three pups, named Romulus, Remus, and Khaleesi, were born on October 1, 2024, through a process involving surrogate domestic dogs. Colossal Biosciences utilized ancient DNA extracted from a 13,000-year-old dire wolf tooth and a 72,000-year-old skull to analyze the species' genome.

By comparing this ancient DNA with the genomes of living canids such as gray wolves, jackals, and foxes, the team identified genetic variants specific to dire wolves. They then employed CRISPR-Cas9 gene-editing technology to modify the DNA of gray wolf cells, introducing 20 changes across 14 genes to replicate dire wolf traits. These modified cells were used to create embryos, which were implanted into the surrogate mothers.


The resulting pups exhibit physical characteristics associated with dire wolves, including broader shoulders, larger bodies, pale coats, and longer, thicker fur. Dire wolves were historically larger than modern gray wolves, with wider heads, stronger jaws, and skulls that could reach up to 12 inches in length, according to the National Park Service. The species was known for hunting megafauna like mastodons and bison, and their extinction is believed to have coincided with the decline of their prey at the end of the Ice Age.

Colossal Biosciences, which has been working on de-extinction projects since 2021, including efforts to revive the woolly mammoth, dodo, and Tasmanian tiger, stated that the dire wolf project had not been previously publicized. The pups are currently housed in a 2,000-acre private facility at an undisclosed location in the northern United States, certified by the American Humane Society and registered with the USDA. The company has expressed long-term plans to potentially restore the species in secure ecological preserves, possibly on indigenous land.

However, the announcement has sparked debate among experts. While Colossal claims this as the world’s first successful de-extinction, independent scientists argue that the pups are not true dire wolves but rather genetically modified gray wolves. Zoologist Philip Seddon from the University of Otago in New Zealand described the animals as "genetically modified grey wolves," noting that dire wolves and gray wolves diverged evolutionarily between 2.5 to 6 million years ago. Dr. Nick Rawlence, also from Otago, emphasized that extinction remains permanent, stating, "Extinction is still forever." Colossal’s chief science officer, Dr. Beth Shapiro, acknowledged that the dire wolves and gray wolves share 99.5% of their DNA, but the company’s goal was to produce animals that could fill the ecological niche left by dire wolves, rather than create an exact genetic replica.

The dire wolf has captured public imagination in recent years, largely due to its depiction in HBO’s Game of Thrones, where the species was portrayed as larger, smarter, and fiercely loyal to the Stark family. George R.R. Martin, author of the A Song of Ice and Fire series on which the show is based, is an investor in Colossal and serves as a cultural adviser. Martin visited the pups and reportedly became emotional, describing the experience as "one of the greatest things that he’s ever seen." He noted the dire wolf’s historical role in the American ecosystem, emphasizing that they are not merely mythical creatures. 

Colossal Biosciences’ CEO, Ben Lamm, called the achievement a "massive milestone," highlighting the potential of their de-extinction technology. The company also revealed that they have created two litters of cloned red wolves, a critically endangered species, as part of their broader conservation efforts. However, questions remain about the ecological role these dire wolf hybrids might play, with some experts, including University at Buffalo biologist Vincent Lynch, cautioning that the technology currently only replicates the exterior traits of extinct species, not their full genetic or behavioral makeup.

As the scientific community continues to debate the implications of this breakthrough, the birth of Romulus, Remus, and Khaleesi represents a bold step into the realm of de-extinction, raising both excitement and ethical concerns about the future of reviving long-lost species.
